Current issues of standardization of antinuclear antibodies’ detection by indirect immunofluorescence on HEp-2 cells

Background. Antinuclear antibodies (ANA) are a group of that target nuclear and cytoplasmic antigens. Testing for ANA using an immunofluorescence assay (IFA) on HЕp-2 cell (IFA-HEp-2) is screening test the diagnosis systemic autoimmune rheumatic diseases, druginduced lupus erythematosus, liver juvenile idiopathic arthritis. In routine testing final report forms bears important, but unstandardized variables (e. g., titers ANA, coding glow types, procedure performing confirmatory tests). Aim study. Develop standard form IFA-HEp-2 results. Material methods . Survey 10 immunological clinical diagnostic laboratories ICAP questionnaire adapted by Working Group Standardization definition method (Committee Immunology Association ‘Federation Laboratory Medicine’). Results. According to results survey: most participants use dilution serum 1:160; indicates maximum end titer antinuclear factor (ANF), staining; identifies staining types (including AC encoding); considers it mandatory make tests detect individual antigens in ANF-positive patients, does not provide list this form; describes ANF each detected glows separately; designates colon give interpretation Based interlaboratory consensus, recommendations requirements ISO 15189, was developed. Conclusions. To achieve high quality interaction between diagnostic/immunological departments health care facilities, it’s necessary introduce into practice results, developed relying modern consensus.
